From Melanotan-2 to Bremelanotide: A Story About One Missing Amide
This article describes published laboratory science for educational purposes. It makes no claims about use in people.
Melanotan-2 is a cyclic heptapeptide: Ac-Nle-cyclo(Asp-His-D-Phe-Arg-Trp-Lys)-NH2. Bremelanotide looks nearly identical, but ends in a free carboxylic acid instead of an amide.
One small difference
| Feature | Melanotan-2 | Bremelanotide |
|---|---|---|
| C-terminus | Amide (-NH2) | Acid (-OH) |
| Mass | about 1,024 Da | about 1,025 Da |
The mass difference is about one dalton, which a mass spectrometer can resolve.
A regulated path
Bremelanotide went through clinical development and was approved by the US FDA in 2019 as a prescription product for a specific condition. That is a regulated pharmaceutical with a defined dose and medical oversight.
