The Melanocortin System and Oil Glands: Why Researchers Study MC5R and Sebum
The melanocortin system is a set of five receptors (MC1R to MC5R) and the hormones that activate them, including alpha-MSH and ACTH. Most people know it for tanning, but it does much more.
A quick receptor map
- MC1R: pigment cells and skin color.
- MC2R: the adrenal glands (stress hormone cortisol).
- MC3R and MC4R: brain circuits of appetite and energy.
- MC5R: found in many tissues, including the sebaceous (oil) glands.
The mouse clue
Mice engineered to lack MC5R show reduced production of the oily secretions of exocrine glands, and studies on human sebocytes (oil gland cells) find that melanocortin signals can influence their lipid production. This is why MC5R is discussed as a possible lever on step one of acne.
Why this is complicated
Melanocortin compounds rarely hit just one receptor. A molecule such as Melanotan-2 10mg acts on several receptors at once, which is exactly why careful researchers compare receptor-selective and non-selective compounds. Melanotan-1 10mg leans more toward MC1R.
The takeaway
Oil glands are under hormonal and peptide control, and melanocortins are one part of that control. It is an active area of basic research with plenty still unknown. It is not a treatment route.
