Endotoxin Testing (LAL) and Research-Grade Peptides: What to Know

Purity and identity are the two numbers most people look for on a peptide report. A third, less visible one is endotoxin. Endotoxins are fragments of bacterial cell walls, and even tiny amounts can disturb sensitive biological experiments.

Key takeaways

  • Endotoxins come from gram-negative bacteria and can survive sterilization.
  • The LAL (Limulus Amebocyte Lysate) assay is the standard test and is reported in endotoxin units (EU).
  • HPLC purity does not measure endotoxin, so it needs its own test.
  • Endotoxin matters most for cell culture and other sensitive assays.

What endotoxin is

Endotoxin is lipopolysaccharide, a component of the outer membrane of gram-negative bacteria. It is heat stable, so autoclaving a solution can kill the bacteria without removing it. Cells in culture, especially immune cells, can respond to very small amounts.

How the LAL test works

The LAL assay uses a clotting reaction from horseshoe crab blood that is triggered by endotoxin. Labs run it in gel-clot, turbidimetric or chromogenic formats. Results are given in endotoxin units per milligram or per milliliter.

Why it is separate from purity

A peptide can be 99% pure by HPLC and still contain trace endotoxin, because endotoxin is not a peptide-related impurity and is present at levels far below what a chromatogram can show.

When it matters for a lab

  • Cell culture work, where endotoxin can change cell behavior
  • Assays involving immune cells
  • Any workflow where a clean negative control is essential
  • Projects that need documented batch quality beyond purity

What to ask a supplier

Ask whether endotoxin testing is available for your lot and what limit was used. If it is not offered, treat that as unknown and design controls accordingly. Our guide on reading a peptide COA covers what a typical report does and does not include.

Common questions

Is endotoxin the same as sterility?

No. Sterility means no living organisms. Endotoxin can remain after organisms are gone.

Do all research peptides come with endotoxin data?

No. It is an extra test, and many suppliers only report purity and identity.
