Lyophilization Explained: Why Research Peptides Ship as Freeze-Dried Powder
Open a vial of a research peptide and you will usually find a small white cake or loose powder. That is the result of lyophilization, also called freeze-drying, and it exists for a good reason: peptides in water do not stay stable for long.
Key takeaways
- Lyophilization removes water by freezing the solution and then sublimating the ice under vacuum.
- Water drives hydrolysis and many other degradation routes, so removing it extends shelf life.
- Freeze-dried peptides are lighter and easier to ship at ambient temperature for short periods.
- The appearance of the cake can vary and is not by itself a quality signal.
The three stages
- Freezing: the peptide solution is cooled until the water turns to ice
- Primary drying: under vacuum and gentle heat, the ice sublimates directly to vapor
- Secondary drying: remaining bound water is driven off at a slightly higher temperature
Why remove the water
Peptide bonds can break in the presence of water, a process called hydrolysis. Water also mobilizes molecules, which speeds oxidation, deamidation and aggregation. By taking the water out, lyophilization slows those reactions by a large factor.
Why the cake looks different from vial to vial
A cake can look fluffy, collapsed, thin or like loose flakes. Fill volume, the presence of bulking agents and the freeze-drying cycle all change the look. A different appearance does not mean a different peptide. Purity and identity come from analytical testing, not from how the powder looks.
Handling a lyophilized vial
Keep it cold and dry. Let the vial reach room temperature before opening, so moisture from the air does not condense on the powder. Our guide to storing lyophilized peptides covers this in more detail.
Common questions
Is freeze-dried always better than liquid?
For storage, almost always. Peptides in solution are far more prone to degradation.
Why is there so little powder visible?
Milligram quantities of peptide form a very small cake. A thin film on the glass is normal.
