Two Letters That Changed a Hormone: Nle4 and D-Phe7 in Melanotan-1
Alpha-MSH is a 13-residue hormone with a short lifetime in biological fluids. Melanotan-1 is the same peptide with just two changes. Those two swaps, one a different amino acid and one a mirror-image version, show how medicinal chemists reshape a natural peptide.
Key takeaways
- Natural alpha-MSH has the sequence Ac-Ser-Tyr-Ser-Met-Glu-His-Phe-Arg-Trp-Gly-Lys-Pro-Val-NH2.
- Melanotan-1 replaces Met4 with norleucine (Nle) and L-Phe7 with D-Phe.
- Norleucine has the same shape as methionine but contains no oxidation-prone sulfur.
- The D-amino acid makes the peptide harder for proteases to cut and raises receptor potency.
- The mass of melanotan-1 is about 1646.8 daltons.
The starting point
Alpha-MSH starts with an acetyl group and ends with an amide, two protections that are also discussed in our terminal modifications post. The core His-Phe-Arg-Trp is the message sequence that activates melanocortin receptors, covered in our melanocortin receptor post. The history is in the Arizona frog-skin story.
Swap one: norleucine for methionine
Methionine contains sulfur that oxidizes easily to methionine sulfoxide, as detailed in our methionine post. Norleucine is a straight four-carbon chain, with a similar length and hydrophobic character but no sulfur. Replacing methionine with it keeps the shape and removes the weak spot.
Swap two: D-phenylalanine at position 7
Almost all natural amino acids are the left-handed L form. Placing the right-handed D form of phenylalanine inside the core changes how the backbone folds, which in the reported work increased potency and lengthened activity. Proteases are built for L residues, so a D residue in the middle also disrupts cleavage.
Why this matters analytically
- The two substitutions change the mass: Nle is 18 lighter than Met, and D-Phe has the same mass as L-Phe, so mass alone cannot detect the D form.
- Telling D from L requires chiral analysis or an HPLC method that separates diastereomers.
- The 1646.8 value should still appear on the spectrum, as discussed in our isotope pattern guide.
- See Melanotan-1 vs Melanotan-2 for how the analogs differ.
Bottom line
Melanotan-1 is a case study in peptide design: replace the fragile residue, mirror the one that matters. For the product and its lot documentation, see our lab reports page.
