The Ring Inside Melanotan-2: How an Asp-Lys Lactam Bridge Makes a Cyclic Peptide
Most peptides are chains. Melanotan-2 is a chain that has been closed into a ring. The ring is not made with the sulfur-sulfur bond many people associate with cyclic peptides, but with a different and sturdier link, called a lactam bridge.
Key takeaways
- Melanotan-2 has the structure Ac-Nle-cyclo(Asp-His-D-Phe-Arg-Trp-Lys)-NH2, a mass of about 1024.2 daltons.
- The ring closes by an amide bond between the side chain of aspartic acid and the side chain of lysine.
- Unlike a disulfide, the lactam is not broken by reducing agents.
- The ring locks the message sequence His-D-Phe-Arg-Trp into a defined shape.
- The same D-phenylalanine trick used in melanotan-1 appears here.
What a lactam bridge is
An aspartic acid side chain ends in a carboxylic acid, and a lysine side chain ends in an amine. A coupling reaction joins them into an amide bond, called a lactam when it forms a ring. In melanotan-2 this links positions 5 and 10 in alpha-MSH numbering and makes a ring of six amino acids. For other ring types, see our cyclic peptide post and the cysteine disulfide story.
Why a ring helps
- Rigidity: the message sequence is held in the shape that fits the receptor, so less energy is wasted on floppy conformations.
- Stability: a ring has no free ends in the cyclic part, and cyclization hides cleavage sites from proteases.
- Reduction resistance: unlike disulfides, a lactam survives reducing conditions in the lab.
- Potency: the original work reported a strong increase over the natural hormone, as in the melanotan-1 post.
Making it
Chemists build the linear chain on a resin, as in our solid-phase synthesis guide, using protecting groups on the Asp and Lys side chains that can be removed selectively. The ring is closed while the peptide is still on the resin. A failed closure leaves linear peptide with a mass 18 higher, from the extra water.
What to look for on a report
- Mass near 1024.2, with no major peak at 1042, which would suggest an open ring.
- A single dominant HPLC peak.
- Acetylation at the start and amidation at the end, both visible in the mass.
- Related impurities, discussed in our impurities post.
Bottom line
The ring makes melanotan-2 a different kind of molecule than a linear peptide of the same length. Check the mass and the cyclization state when reading its report; see the lab reports page and the MT-1 vs MT-2 comparison.
